摘要 目的调查分析地高辛血药浓度的测定方法、血药浓度的影响因素,并建立用高效液相色谱法测定地高辛血药浓度的方法。方法采用沃特世ACQUITYUPLC超高效液相色谱仪,沃特世XSELECTCSHC18(4.6×250mm,5μm),流动相:甲醇︰水(65︰35);流速:1.0mL/min,检测波长:220nm,柱温30℃,进样量:20μL。结果地高辛血药浓度在0.5~3.0ng/mL时,与峰面积呈良好的线性关系(r=0.9978);该方法回收率较高,RSD为1.59%~3.13%。结论高效液相色谱法测定地高辛血药浓度可靠、准确、灵敏度高,可用于地高辛血药浓度测定。 Objective Investigation of the digoxin plasma concentration, plasma concentration of influencing factors, to establish the method of determining the blood concentration of Digoxin. Methods Waters ACQUITY UPLC; Waters XSELECT CSH C18 ( 4.6 × 250 mm,5μm ) ; mobile phase: methanol : water (65 : 35); flow rate: of 1.0 mL/mL, Detection wavelength: 220hm, column temperature 30℃, into the sample volume: 20 μL. Results A good linearity between the concentrations of the samples and their peak alias was shown from 0.5-3.0 ng/mL(r=0.9978). This method has the high recovery, RSD=1.59%-3.13%. Conclusion High performance liquid chromatography determination to digoxin blood concentration is reliable, accurate, high sensitivity, may be used to to digoxin plasma concentration.
