摘要
目的通过对去卵巢小鼠补充雌二醇及黄体酮治疗,观察外周血Th1/Th2细胞因子水平变化,探讨雌二醇及孕酮对外周血Th1/Th2细胞因子水平的影响。方法将80只健康雌性小鼠随机分为4组,每组20只,假手术组不给药物,3组去卵巢小鼠分别接受苯甲酸雌二醇、黄体酮及注射油皮下注射治疗。每周2次,共注射25次。采用ELISA法检测4组血清雌二醇、孕酮、转化生长因子(TGF—β)、肿瘤坏死因子-CC(TNF—α)、白细胞介素2(IL-2)、白细胞介素6(IL-6)、白细胞介素10(1L-10)、卵泡刺激素和黄体生成素水平。结果(1)与假手术组比较,术后12周非用药组血清TNF—α、IL-6水平明显升高(均P〈005),IL-10水平降低(P〈0.05),IL-2、TGF—β水平无明显改变(P〉0.05)。(2)雌二醇组与非用药组比较,血清TNF-α、IL-6水平明显降低(P〈005或0.01),而IL-10水平显著升高(P〈0.05)。(3)黄体酮组与非用药组比较,TNF—α水平降低,而IL-10水平升高,差异有统计学意义(均P〈0.05)。结论小鼠去卵巢后机体免疫反应倾向Th1型,生理浓度雌二醇及孕酮均可推动机体免疫反应向Th2型转变。
Objective To investigate the effects of estradiol(E2) and progesterone(P) on peripheral blood Th1/Th2 in ovariectomized mice. Methods Total 80 female mice were divided into 4 groups at random: in INT group mice received sham operation, in OVX+EB group ovariectomized mice treated with estradiol, in OVX+P group ovariectomized mice treated with progesterone, in OVX group ovvariectomized mice treated with oil. Injections were performed twice per week with 25 times in total. The serum levels of P, E2, transforming growth factor(TGF-15 ), interleukin-2(IL-2), tumor necrosis factor-α (TNF-α), interleukin-6(IL-6), interleukin-10(IL-10), follicle-stimulation hormone(FSH), luteinizing hormone (LH) were measured by enzyme linked immunoserbent assay (ELISA). Results Compared with INT group the levels of TNF-α, IL-6 in OVX group were significantly higher(P〈0.05), while the levels of IL-10 were lower(P〈0.05). There was no difference in IL-2, TGF- 13 levels between two group. The levels of TNF-α, IL-6 in OVX+EB group were significantly lower than those in OVX group(P〈0.05 or 0.01),while IL-10 levels were significantly higher(P〈0.05). OVX+P group had higher IL-10 and lower TNF-α levels when compared with the OVX group (P〈0.05). Conclusion In ovariectomized mice, a Thl-type response is developed ; the biological concentration of E2 and P may switch organ immune response to Th2-type.
