摘要
为了对流浪猫粪便中分离的贾第虫滋养体进行基因型鉴定,提取虫体基因组DNA,以16SrRNA和β-贾第素基因为遗传标记,通过PCR扩增、克隆、测序、NCBI比对以及相关软件分析,建立系统进化树,确定了其基因型。结果显示,成功扩增出291bp的16SrRNA和511bp的β-贾第素基因片段,测定了2个基因片段的序列,并经NCBI比对以及DNAStar中MegAlign软件分析确定该株贾第虫为十二指肠贾第虫F型。本研究首次对我国猫源贾第虫进行了基因型鉴定,为猫源贾第虫分子生物学和分子流行病学研究奠定了基础。
The present study aimed at genotyping the trophozoites of Giardia sp.that was firstly isolated from stray cat feces of Guangzhou,China.The genomic DNA was extracted from fecal sample and two loci of 16 S rRNA gene and β-giardin gene were used as gene markers for genotyping.After PCR amplification,two fragments of 16 S rRNA(291 bp) and β-giardin gene(511 bp) were obtained and sequenced.The phylogenetic trees were established at 16 S rRNA and β-giardin loci by MegAlign of DNAStar software.Results showed that the cat-derived Giardia sp.was Giardia duodenalis and belonged to assemblage F.The results laid a foundation for the molecular biology and molecular epidemiology studies of cat-derived Giardia in China.
