HIWI在食管癌细胞系Eca-109中的表达及意义 被引量:2

摘要 目的检测干细胞基因HIWI在食管癌细胞系Eca-109中的表达及其对细胞增殖的影响。方法通过细胞免疫荧光检测食管癌细胞系Eca-109中HIWI蛋白的表达;应用LipofectamineTM2000将HIWI特异的siRNA转染进细胞中,再通过RT-PCR检测siRNA的沉默效率;通过MTT检测转染后各转染组细胞的生长曲线,观察HIWI对食管癌细胞增殖的影响。结果 HIWI蛋白在食管癌细胞系Eca-109中高表达,且细胞之间存在差异;HIWI特异的siRNA沉默效率可高达80%左右;转染测得HIWI特异siRNA组较各对照组细胞的增殖速度减慢并有统计学意义。结论 HIWI在食管癌细胞系Eca-109中高表达,细胞中存在食管癌干细胞;降低HIWI的表达后能够减慢食管癌细胞的增殖速度。
