摘要
目的建立测定芪胶膏中黄芪甲苷含量的高效液相色谱法。方法采用Waters Symmetry Shield TM RP色谱柱C18(5μm 4.6 mm×250 mm,柱子编号:186000112),以乙腈-水(34∶66)为流动相,流速1.0 ml/min,柱温40℃,漂移管温度60℃,载气压力30 psi。结果黄芪甲苷进样量在1.9046~13.9674μg范围内时,与峰面积积分值有良好的线性关系(r=0.9991,n=6),平均加样回收率为98.52%,RSD=3.7%(n=6)。结论高效液相色谱-蒸发光散射检测法灵敏、简便、重复性好,可用于芪胶膏的质量控制。
Objective To establish the high performance liquid chromatogram - evaporative light scattering detection ( HPLC - ELSD) method to determine the content of astragaloside IV in Qijiao Paste. Methods The conditions for detection of astragaloside IV content by HPLC were Waters Symmetry Shield TM RP chromatographic column C18 (5 μm, 4.6 mm × 250 mm, column NO: 186000112), acetonitrile to water (34: 66) used as mobile phase with the velocity of 1.0 ml/min, column temperature of 40℃, drift tube temperature of 60 ℃, and carrier gas pressure of 30 psi. Results When the concentration of astragaloside IV in the sample was within the range of 1. 9046 - 13. 9674 μg, it had a good linear relation with peak area score ( r = 0. 9991, n = 6). The average sample recovery rate was 98.52%, and RSD was 3.7% ( n = 6). Conclusion This method is sensitive, simple, and reproducible, which can be used for the quality comtrol of Qijiao Paste.
