大黄素对非酒精性脂肪肝小鼠肝脂质沉积的作用及机制 被引量:16

Effects and mechanism of emodin on treatment of non-alcoholic fatty liver disease

摘要 目的:研究大黄素改善非酒精性脂肪肝小鼠肝脂质沉积作用及分子机制。方法:建立非酒精性脂肪肝的C57BL6小鼠模型,随机分成模型组、大黄素组、罗格列酮组,空白对照组为正常喂养小鼠,每组5只。治疗4周后测定4组动物治疗前后的体质量变化以及血甘油三酯水平,比较4组动物治疗前后的肝脏湿重和病理变化,同时使用Western Blot方法检测脂联素受体2的表达情况。结果:与模型组比较,罗格列酮组治疗后的体质量、肝脏湿重、肝指数均下降,且大黄素组甘油三酯水平均明显降低(P<0.05);肝组织脂质沉积明显减轻;脂联素受体2表达显著升高。结论:大黄素对于非酒精性脂肪肝脂质沉积具有较好改善作用,其作用机制与增加脂联素受体2的表达有关。 Objective: To explore the effects of Emodin on treatment of non-alcoholic fatty liver disease in mouse. Methods: C57BL6 mouse of non-alcoholic fatty liver model were randomly divided into three groups: untreated group, emodin group and rosiglitazone group and were treated with DMSO, emodin and rosiglitazone by intraperitoneal injection respectively. Every group has 5 mice. Another 5 mice administrated by normal diet were used as control group. After 4-weeks treatment, the following indexes were detected: the rat's body weights, triglyceride in blood, the wet weight of the liver in each group. The specimen of liver was collected to undergo pathological examination. Western blotting was used to detect the protein expression of adiponectin receptor 2. Results: Compared with the untreated group, the body weight, liver wet weight and liver index of the mouse in emodin group decreased, and blood triglyceride levels were remarkably decreased (P〈0.05). The pathological changes of the liver were remarkably milder and the adiponctin receptor 2 remarkably increased in emodin group. Conclusion: Emodin is effective in treatment of non-alcoholic fatty liver disease in mouse. Increasing the content of adiponctin receptor 2 in the liver may be the potential mechanism.
