Comparative study of methods for isolation of total DNA from Schisandra chinensis

摘要 目的:建立一套适合从五味子种子中提取高质量DNA的方法,为进一步开展五味子分子鉴定奠定基础。方法:比较SDS法、高盐低pH值法以及改进的CTAB法,运用紫外光谱和电泳分析方法测定所得DNA样品的纯度与浓度。结果:3种方法中CTAB改良法所得DNA纯度最高,完整性好,且该方法稳定可靠。结论:CTAB改良法是适合从五味子中提取高质量DNA的方法。 Objective: To obtain a method for yielding DNA from Schisandra chinensis, which lie a foundation for further molecular experiments. Methods: Three different DNA extraction protocols, SDS protocol, high salt with low pH protocols and the improved CTAB protocol, were examined and compared. The purity and concentration of DNA were detected by UV spectrophotometric analysising and agarose gel electrophoresis testing. Results: The DNA isolated by improved CTAB protocol was with least impurity in three different DNA extraction protocols. Conclusion: The improved CTAB protocol was suitable for the DNA isolation from Schisandra chinensis. The DNA isolated by this protocol could be used in the further molecular experiments.
