摘要
p66Shc是调控细胞氧化应激和生命周期的关键蛋白,在细胞氧化损伤过程中发挥重要作用。糖尿病肾病是引起终末期肾疾病的主要原因,细胞活性氧的过度产生是糖尿病肾病发生和进展的核心环节。本文结合近期p66Shc研究现状和作者以往的研究成果,简要介绍p66Shc基因结构与功能、转录调控、蛋白修饰及线粒体转位及其在与细胞氧化损伤的关系,以及p66Shc基因在糖尿病肾病发病中的作用。
p66Shc,an adaptor protein,is known as a key regulator of cellular oxidative stress and life cycle,eventually leading to cellular oxidative injury.Diabetic nephropathy is a major cause of end-stage renal disease.Over-production of reactive oxygen species is regarded as the key process in the pathogenesis of diabetic nephropathy.In this review,we summarize the structure,function,transcription,post-translational modification to the p66Shc protein,and mitochondrial dysfunction of p66Shc gene,as well as the physiological relationships between mitochondrial dysfunction of p66Shc and reactive oxygen species,and the role of p66Shc in the pathogenesis of diabetic nephropathy.
