摘要
目的探讨大鼠急性胰腺炎(AP)早期细胞因子和胰腺细胞凋亡的动态变化。方法将40只SD雄性大鼠随机分为正常对照(NC)组和急性胰腺炎(AP)组。采用经腹腔单次注射L-精氨酸制作大鼠急性胰腺炎模型。各组分别于造模后24 h、36 h处死半数大鼠,观察血清淀粉酶、细胞因子IL-1β、TNF-α和IL-10的变化。并对胰腺病理学变化、胰腺细胞凋亡情况定位评分。结果造模后24 h,AP组血淀粉酶和IL-1β、TNF-α均较NC组升高(P<0.01),而IL-10与NC组相当;AP组胰腺细胞凋亡指数较NC组增加(P<0.01),胰腺组织损害明显增强(P<0.01)。造模后36 h,AP组血淀粉酶水平下降,IL-1β、TNF-α及IL-10水平继续上升,胰腺病理损害进一步加重(P<0.01)。同时,AP组凋亡指数下降,但仍高于NC组(P<0.01)。结论作为保护性机制,在AP发生时胰腺细胞凋亡明显增加;但这种内源性凋亡的增加出现早,维持时间短,不足以抑制强烈的炎症反应。
Objective It is to approach the dynamic changes of cytokines and apoptosis of pancreatic cells in acute pancre- atitis (AP) rat. Methods Forty SD male rats were randomly divided into normal control group (NC) and AP group with 20 rates in each one. The AP model was made by single intraperitoneal injection of L - arginine. Half of animals in each group were killed at 24 - and 36 - hour after injection, the changes of serum amylase, interleukin (IL) - 1β, tumor necrosis factor (TNF) -α and IL- 10 were observed, and the index of pancreatic cell apoptosis and histological were evaluated. Results At the first 24 - hour in AP group, the levels of amylase, IL - 1β and TNF - α were higher than those in NC group ( P 〈 0. Ol ) , but the level of IL - 10 was as the same as in NC group. The index of pancreatic cell apoptosis and the pancreatic damage level in AP group were higher than those in NC group (P 〈0.01 ). After 36 -hour, the level of amylase in AP group went down, but the level of IL - 1β,TNF - α and IL - 10 continued to increase and the pancreatic damage aggravated. The index of pan- creatic cell apoptosis decreased in AP group, but also higher than that in NC group (P 〈 0.01 ). Conclusion As a protective mechanism, apoptosis in pancreatic cell can significantly increase in AP. But the increasing of pancreatic apoptosis is not only early but transitory and it could not inhibit the strong inflammatory response.
