摘要
目的测定药物制剂和血浆中的β-内酰胺酶抑制剂舒巴坦的含量。方法使用一种灵敏、简单、廉价的流动注射化学发光法,这种方法的原理是,在碱性环境中,舒巴坦对鲁米诺和过氧醋酸发光反应有抑制作用。结果在适当的条件下,检测限为2.0ng·mL-1(3σ),舒巴坦的浓度为5.0ng·mL-1至1 000ng·mL-1的范围内,发光的强度与舒巴坦的浓度成线性关系。回归方程为:ΔI=90.195 C+19.54(ΔI表示发光强度,C表示舒巴坦浓度,μg·mL-1),相关系数r2=0.9975。结论这种方法已经成功地应用于2种药物制剂和血浆中舒巴坦的测定。
Aim To determine the beta-lactamase inhibitors Sulbactam in pharmaceutical preparations and added to blood serum.Methods By using a sensitive,simple and inexpensive flow injection chemiluminescence method.The method is based on the inhibitory effect of Sulbactam on the chemiluminescence reaction between luminol and peroxy-acetic acid in alkaline medium.Results Under the optimal conditions,the decrement of chemiluminescence intensity was linear with Sulbactam concentration over the range from 5.0 ng·mL-1 to 1 000 ng·mL-1 with the detection limit of 2.0 ng·mL-1(3σ).The regression equation was ΔI=90.195C+19.54(ΔI is the relative CL intensity and C is the concentration of Sulbactam,μg·mL-1) with a correlation coefficient r2=0.997 5.Conclusion The method has been successfully applied to the determination of Sulbactam in two kinds of pharmaceutical preparations and serum samples.
