聚合酶链反应结合斑点杂交测定乙型肝炎病毒DNA的评价

Evaluation or Polymerase Chain Reaction and Dot blot for HBV DNA Detection

分别以Ｄｏｔｂｌｏｔ，ＰＣＲ－ＥＢ，ＰＣＲ－Ｄｏｔｂｌｏｔ法测定了１４６例慢性乙型肝炎患者血清ＨＢＶＤＮＡ。结果显示在ＨＢｅＡｇ组，ＰＣＲ－Ｄｏｔｂｌｏｔ阳性率（１００％）显著高于ＰＣＲ－ＥＢ（９４．８％），在ＨＢｅＡｂ组，ＰＣＲ－Ｄｏｔｂｌｏｔ的检出率为５４％，ＰＣＲ－ＥＢ为３８％。两者总检出率分别为８４．２％和７５．３％，统计学处理差异显著。有３例ＰＣＲ－ＥＢ再现生信号。

Serum HBV DNA in 146 cases of patients with chronic HBV infection was detected by means of Dot blot, PCR-EB and PCR-Dot blot methods. The results showed that the positive rates of PCR-Dot blot (100%) in the group of HBeAg+were higher than that of PCR-EB (94.8%) (P<0.05). In the group of HBeAb+, the positive rates of PCR-Dot blot and PCR-EB were 54% and 38%. The total positive rates were 84.2% and 75.3%. There was a markedly difference between them (P<0.05). 3 cases had PCR-EB positive signals, but PCR-Dot blot testing was negative. The results suggested that PCR-Dot blot may be far more sensitive and have good specificity.
