摘要
目的 :观察自由基对培养的人胚软骨细胞损伤及硒的防护作用。方法 :在体外培养的人胚关节软骨细胞培养液中 ,加入黄嘌呤、黄嘌呤氧化酶及微量元素硒。测定软骨细胞DNA和蛋白质及软骨基质中蛋白多糖和胶原的含量。结果 :培养 14 4h后 ,软骨细胞DNA、蛋白质及软骨基质中蛋白多糖含量都明显减少 ,但胶原含量无明显改变 ,含硒组减少程度轻。结论 :自由基可能和退形性关节疾病病因有关 ,是否选择微量元素硒预防及治疗自由基引起的软骨损伤需动物实验的进一步研究。
Objective: To observe the damage of cultured human embryo chondrocytes by free radicals and the reversed effect of selenium.Method:Xanthine xanthine oxidase and trace element selenium were put into the human embryo articular chondrocytes culture liquid in vitro.The syntheses of DNA,protein,proteoglycan and collagen were measured.Results:After the involvement of xanthine and xanthine oxidase for 144 hours,free radicals could inhibit significantly the syntheses of DNA,protein and proteoglycan,but not collagen.Selenium could reverse the effect of free radicals on chondrocytes to some degree.Conclusion:Free radicals might be related to the pathogenesis of degenerative joint disease. Whether selecting selenium to reverse the cartilage damage by free radicals needs further study on animal experiment.
