摘要
目的 :探讨儿童自身免疫甲状腺病 (AITD)细胞因子失衡与T细胞亚群失衡的关系。方法 :采用E花环法和ELISA法同步分别检测初诊的AITD患儿和正常儿童外周血T细胞亚群和单个核细胞在丝裂原诱导下产生的IFN -γ、IL - 4和IFN -γ IL - 4比值。结果 :与正常组相比 ,CLT和GD病儿IFN -γ水平和IFN -γ IL - 4比值明显升高 ,IL - 4水平明显降低 ,CD+3、CD+8明显降低 ,CD+4 CD+8比值明显升高。且CD+8与IL - 4水平、CD+4 CD+8比值与IFN -γ IL - 4比值呈显著正相关 (r分别为 0 .76 0 .75 ,0 .6 8 0 .5 8,P <0 .0 5 )。结论 :AITD患儿Th1类细胞因子升高可能主要是CD+4Th1亚类的功能亢进所致 。
Objective: To explore the relationship between the imbalance of cytokinesis and T cell subsets in children with autoimmune thyroid disease. Methods: IFN-γ, IL-4 and the ratio of IFN-γ/IL-4 released from stimulated peripheral blood mononuclear cells (PBMC) and their relationship with T cell subsets were investigated in 22 children with Hashimoto’s thyroiditis (HT), 24 children with Graves’disease (GD) at presentation. Results: Compared with normal subjects, IFN-γ, the ratio of IFN-γ/IL-4 were significantly increased whereas IL-4 significantly decreased in children with HT and GD at presentation. The percentage of CD + 3, CD + 8 T cells in peripheral blood were lower but the ratio of CD + 4/CD + 8 was higher in children with HT and GD at presentation than those in normal controls. The ratio of CD + 4/CD + 8, the percentage of CD + 8 T were positively correlated with ratio of IFN-γ/IL-4 and IL-4 respectively. Conclusions: It is possible that the increased Th1 type cytokinesis be attributed to the enhanced function of Th1 type CD + 4 cells while the decreased Th2 type cytokinesis be attributed to the declined number and function of Th2 type CD + 8 cell.
