摘要
目的观察缪刺电针和患侧电针足三里穴、阳陵泉穴对坐骨神经慢性缩窄性损伤(CCI)模型大鼠痛阈和背根神经节P2X3受体表达的影响。方法通过机械刺激法和热辐射法测定大鼠机械缩足反射阈值(MWT)和热缩足反射潜伏期(TWL),采用Western blotting法观察大鼠造模侧L4-L6DRG上P2X3受体蛋白的表达。结果①与假模组比较,CCI术后各组大鼠在各时间点MWT和TWL明显降低(P均<0.001);电针7天后,与模型对照组相比,缪刺电针组和患侧电针组MWT和TWL升高(P均<0.05);缪刺电针组和患侧电针组MWT和TWL差异无统计学意义(P均>0.05);②CCI术后第14天,与假模组相比,各组大鼠P2X3受体蛋白表达上调(P均<0.05),与模型对照组相比,电针能够明显减少P2X3受体蛋白的表达(P均<0.001),缪刺电针组与患侧电针组相比,差异没有显著性(P>0.05)。结论缪刺电针和患侧电针足三里穴、阳陵泉穴,均可下调CCI大鼠DRG神经元中P2X3受体蛋白的表达,减轻CCI大鼠机械性痛觉过敏和热痛觉过敏。
Objective To observe the chang of P2X3receptor protein expression located in the L4 to L6 segments of dorsal root gan- glion(DRG) by electroacupuncture (EA) Zusanli acupoint (ST36) and Yanglingquan acupoint (GB34) on rats with chronic constrictive injury(CCI) of the right sciatic. Methods The mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were measured by mechanical stimulation method and thermal radiation electronic. The expression of the P2X3 receptor protein was detected by Western blotting. Results (!)After CCI operation,the MWT and TWL of each group rat were significantly decreased(P 〈 O. 001 ) as com- pared to the sham group on every post -operative day. In the contralateral EA group and ipsilateral EA group, MWT and TWL were in- creased (P 〈 0. 05 ) compared with the model control group after EA treatment 7 days. There was no significant difference between the con- tralateral EA group and ipsilateral EA group ( P 〉 0.05 ). (~)After CCI operation 14 days, there was increased expression of P2X3 receptor protein in each group compared with the sham group( P 〈 0.05 ). After electroacupuneture treatment 7 days, the P2X3 receptor protein ex- pression was decreased at a different degree in the contralateral EA group and ipsilateral EA group, and the changes were significant differ- ences compared with the model control group ( P 〈 0. 001 ). But there was no significant difference between the contralateral EA group and ipsilateral EA group (P 〉 0.05). Conclusion Electroacupuncturing eontralateral and ipsilateral Zusanli acupoint and Yanglingquan acu- point can down - regulate the protein expression of P2X3 receptor in the L4 - L6 DRG at operation side,and can alleviate mechanical hy- peralgesia and thermal hyperalgesia induced by CCI.
