摘要
目的应用柱切换高效液相色谱技术实现鼠尾草属植物中水溶性和脂溶性化学成分在不同色谱柱上的分析,实现更多成分的分离。方法在HPLC基础上利用二位十通切换阀实现水溶性成分和脂溶性成分的切割,以Megres-C18色谱柱(4.6 mm×10 mm,5μm)为预吸附柱,用Zorbax Sb-Aq色谱柱(4.6 mm×150 mm,5μm)完成水溶性成分分析,用Eclips XDB-C18色谱柱(4.6 mm×150 mm,5μm)完成脂溶性成分分析。结果建立了柱切换高效液相色谱技术分离鼠尾草属植物化学成分的方法,完成其中7种成分(丹参素、原儿茶醛、原儿茶酸、咖啡酸、隐丹参酮、丹参酮I和丹参酮IIA)的定性和定量分析。结论利用柱切换高效液相色谱技术可以很好地实现鼠尾草植物中极性差异较大化学成分的分离和分析,为复杂中药的质量控制提供了一种方法借鉴。
AIM To analyze the hydrophilic and hydrophobic components of Salvia miltiorrhiza,Salvia yunnanensis,Salvia przewalskii and Salvia castanea and to achieve the separation of much more components with the HPLC with column switching technique.METHODS Adding a two positions ten-pass switching valve to achieve the cutting of the water soluble and fat-soluble chemical compositions in the use of Megres-C18 column(4.6 mm×10 mm,5 μm) as a pre-column,the hydrophilic constituents and the hydrophobic constituents were separated on a Zorbax Sb-Aq column(4.6 mm× 150 mm,5 μm) and an Eclips XDB-C18 column(4.6 mm× 150 mm,5 μm).RESULTS An HPLC method coupled with column switching was successfully applied to the analysis of hydrophilic and hydrophobic components in four Salvia plants.Seven active components(danshensu,protocatechuic aldehyde,protocatechuic acid,caffeic acid,cryptotanshinone,tanshinone I and tanshinone IIA) in these plants were qualitatively and quantitatively analyzed.CONCLUSION Data on HPLC with column switching shows that the separation and analysis of the different polarity components of the Salvia can be achieved.
