摘要
目的观察三丫苦对高脂饮食性胰岛素抵抗大鼠脂联素、瘦素和抵抗素的影响。方法将40只SD雄性大鼠随机分为4组:正常对照组、模型组、三丫苦组、罗格列酮组,建立高脂饮食性胰岛素抵抗大鼠模型。检测各组大鼠TC、TG、FBG、FINS等指标的变化,清醒状态下高胰岛素-正葡萄糖钳夹实验检测葡萄糖输注率,ELISA法检测血清脂联素、瘦素和抵抗素水平。结果与正常对照组比较,模型组TC、TG、FBG、FINS均升高(P<0.01),葡萄糖输注率降低(P<0.01)。给予三丫苦后,大鼠TC、TG、FBG、FINS降低(P<0.01或P<0.05),GIR水平升高(P<0.05),与罗格列酮组比较,TC和TG降低(P<0.05)。与正常对照组比较,模型组APN和LP水平均显著降低(P<0.01),抵抗素水平显著升高(P<0.01)。与模型组比较,三丫苦组和罗格列酮组APN和LP水平均有不同程度的降低(P<0.01或P<0.05),三丫苦组抵抗素水平降低(P<0.01)。与罗格列酮组比较,三丫苦组抵抗素水平降低(P<0.05)。结论三丫苦对高脂饮食性胰岛素抵抗大鼠脂联素、瘦素和抵抗素有一定调节作用。
Objective To observe the evodia lepta's effect on serum adiponectin, leptin and resistin in insulin resistent (IR) Rats of fat-rich diet. Methods Forty male SD rats were randomly divided into control groups, model group, evedia lepta treating group, rosiglitazone treating group, and the insulin resistent (IR)rats model with fat-rich diet were estab-lished. The changes of total cholesterol( TC), tatal glycerin (GC), fasting blood glucose ( FBG), fasting insulin (Fins) of all rats were measured. In an awake stste, the glucose infusion rates were detected by experiment of hyperinsulinemic - euglycemic clamp, while the levels of serum adiponectin, leptin and resistin were tested by ELLSA. Results Compared with control group, the level of TC, TG, FBG, FINS in model group were increased( P 〈0.01 ), also the glucose infu-sion rates were decreased( P 〈0.01 ). After treating by evodia lepta, the level of TC, TG, FBG, FINS were decreased (P 〈 O. 01 orP 〈0.05) and the GIR level was increased, while compared to rosiglitazone treating group, the level of TC, TG were decreased. The APN and LP of model group were decreased significantly than that of control group (P 〈 0.01 ), but on the other the resistin was significantly increase( P 〈0.01 ). Comparing with model group, the level of APN and LP in both evodia lepta treating group and rosiglitazone treating group were reduced to varying degrees (P 〈 O. O1 or P 〈0.05 ), at same time, the resistin in evodia lepta treating group was reduced( P 〈0. 01 ). Compared with rosiglitazone treating group, the level of resistin in evodia lepta treating group was decreased ( P 〈 0. 05 ). Conclusions The regulation effect of evodia lepta on serum adiponectin, leptin and resistin in IR rats of fat - rich diet was observed to varying degrees.
