摘要
目的肌注甘油复制急性肾功能衰竭(ARF)兔模型,观察葛根素(Pue)对ARF兔血液流变学和肾血流量的影响。方法健康雄性日本大耳白兔30只,随机分为正常组、ARF模型组、Pue1组(20mg/kg)、Pue2组(40mg/kg)、Pue3组(80mg/kg)。各组于不同时间点测量其肾血流量、血液流变学指标和肾功能指标(Cr、BUN),并观察肾组织形态学改变。结果与模型组比较,Pue2组和Pue3组治疗后各时间点Cr、BUN降低(P<0.05),血液流变学指标降低明显(P<0.05),肾血流量增加差异具有统计学意义(P<0.05)。Pue2组和Pue3组肾小管上皮细胞肿胀减轻,管型少见。结论葛根素可明显改善急性肾功能衰竭兔血液流变性,增加肾血流量,进而达到改善、减轻肾小管损害的作用。
Objective To investigate the effects of puerarin on hemorheology and renal hemodynamics in rabbits with glycerine-acute renal failure. Methods 30 male rabbits were used and divided into 5 groups randomly : ( 1 ) Normal group ; (2) ARF group, acute renal failure induced by glycerine; (3) Pue 1 group, inject puerarin intravenously in a dose of 20 mg/kg after ARF ; (4) Pue 2 group, puerarin treated in a dose of 40 mg/kg after ARF ; (5) Pue 3 group, puerarin treated in a dose of 80 mg/kg after ARF. The following indices of each group were measured at various times, including hemorheolo- gy, renal blood flow (RBF), blood urea nitrogen (BUN) and serum creatinine levels (Cr). At last, left kidneys were har- vested for histopathological examination. Results Pue 2 group and Pue 3 group showed significantly decreased level of he- morheology, as well as Cr and BUN and increased level of RBF compared with ARF group ( P 〈 0.05 ). Histopathological examination showed that the swelling of renal tubular epithelial cell was lightened and cast formation was reduced obviously. Conclusion Puerarin would improve hemodynamics and increase renal blood flow in order to improve renal function and re- duce tubular injury.
