去氢甲睾酮ELISA试剂盒的研制

摘要 以实验室自主研发的一株能稳定分泌去氢甲睾酮单克隆抗体的杂交瘤细胞株为基础,应用ELISA原理研制去氢甲睾酮残留快速检测试剂盒,并对试剂盒特性进行测定。结果表明,该试剂盒标准曲线的回归方程为y=-0.205 5x+0.516(R2=0.993 1),线性检测范围为0.1ng/mL~100ng/mL,半数抑制浓度(IC50)为3.20ng/mL,对猪肉加标样品的检测回收率在87.41%±5.44%~110.70%±2.05%之间,试剂盒检测与去氢甲睾酮结构类似物丙酸睾酮酯、群勃龙的交叉反应率均小于1%;试剂盒在4℃能稳定保存6个月。说明试剂盒能够应用于食品中残留去氢甲睾酮的快速检测。 A sensitive and specific monoclonal antibody(mAb)-based enzyme-linked immunosorbnent assay(ELISA) kit for detecting DMT(1-dehydro-17a-methyltestosterone)was developed,and the performance of the kit was also characterized in this paper.The linear range of the calibration curve was 0.1 to 100 ng/mL;its half inhibitory concentration(IC50) reached 3.20 ng/mL;the recoveries of meat samples varied from 87.41%±5.44% to 110.70%±2.05%;its cross-reactivity value with structural analogues TP and TB were less than 1%.The developed kit still remained effective after six month storage at 4 ℃.These results indicated that the established ELISA kit has a potential application to rapid determination of DMT residue in food samples.
