清养透解法对自发性红斑狼疮模型小鼠血管损伤因子的影响 被引量:1

Effect of Therapy of Clearing Heat,Nourishing Yin and Expelling Pathogens on Vascular Injury Factor in Spontaneous Systemic Lupus Erythematosus Mice

摘要 【目的】观察清养透解法干预自发性红斑狼疮模型(MRL/lpr)小鼠血管损伤的作用。【方法】将24只8~10周雌性MRL/lpr小鼠按体质量随机分为中药组、西药组、中西医结合组、模型对照组4组,每组6只,另取8~10周龄雌性BALB/c小鼠6只为正常对照组。中药组予以清养透解合剂灌胃(剂量为9.5 g.kg-1.d-1),西药组予以强的松灌胃(剂量为10 mg.kg-1.d-1),中西医结合组予以强的松并清养透解合剂灌胃,模型对照组予以生理盐水灌胃,连续给药6周,正常对照组则不予任何处理。6周后取血做血栓调节蛋白(sTM)、血管假性血友病因子(vWF)检测,并取小鼠肾脏在透射电子显微镜下观察其病理变化。【结果】与正常对照组比较,模型对照组血清sTM、vWF水平显著上升(P<0.01);与模型对照组比较,中药组、中西医结合组可显著降低血清sTM、vWF水平,西药组可显著降低vWF水平(P<0.05或P<0.01);西药组、中西医结合组均能改善MRL/lpr小鼠肾组织病理改变。【结论】清养透解合剂能降低系统性红斑狼疮模型小鼠血清vWF、sTM的表达,保护血管内皮,改善肾组织慢性炎症病理改变。 Objective To observe the effect of therapy of clearing heat, nourishing yin and expelling pathogens on vascular injury in spontaneous systemic lupus erythematosus MRL/lpr mice. Methods Twenty-four female MRL/lpr mice aged 8-10 weeks were equally randomized into 4 groups, i. e. , Chinese medicine group, western medicine group, Chinese and western medicine group, and model group. Six female healthy BALB/C mice aged 8-10 weeks were in the normal control group. Chinese medicine group was given gastric gavage of herbal mixture with the actions of clearing heat, nourishing yin and expelling pathogens (9. 5 g ·kg^-1·d^-1), western medicine group was given prednisone ( 10 mg ·kg^-1·d^-1 ), Chinese and western medicine group was given the herbal mixture and prednisone, and the model group was given normal saline. The treatment lasted 6 weeks. No medication was given for the normal control group. Six weeks later, serum soluble thrombomodulin (sTM) and yon Willebrand factor (vWF) levels were detected, and the renal pathological changes in mice were also observed under transmission electron microscope. Results Serum sTM and vWF levels were higher in the model group than those in the normal control group (P 〈 0. 01 ). Compared to the model group, sTM and vWF levels were decreased in the medication groups, and the decrease of vWF was obvious in western medicine group ( P 〈 0. 05 or P 〈 0. 01 ). The renal pathological changes were relieved to various degrees in the three medication groups. Conclusion The therapy of clearing heat, nourishing yin and expelling pathogens shows certain protective effect on vascular endothelia by decreasing serum sTM and vWF levels, and on relieving the renal pathological changes of chronic inflammation in MRL/lpr mice.
