摘要
目的观察糖心乐对糖尿病心肌病大鼠血糖和心肌酶的影响。方法 SD大鼠腹腔注射链脲佐菌素建立糖尿病模型,成模后分为模型对照组、格列齐特对照组及糖心乐大、小剂量组,每组12只。造模后8周格列齐特对照组给予格列齐特20 mg·kg-1灌胃治疗,糖心乐大、小剂量组分别给予2.2 kg.L-1糖心乐和1.1 kg.L-1糖心乐6.5 mL.kg-1灌胃治疗,模型对照组灌服生理盐水,每日1次,连续4周。另取10只大鼠为正常对照组,不予任何处理。测定灌胃治疗4周后各组大鼠的空腹血糖(FBS)和心肌酶活性。结果治疗4周后,与正常对照组比较,模型对照组、格列齐特对照组及糖心乐大、小剂量组大鼠FBS明显升高(P<0.01),心肌肌酸激酶(CK)和乳酸脱氢酶(LDH)含量明显降低(P<0.01);与模型对照组比较,格列齐特对照组和糖心乐大剂量组大鼠FBS明显降低(P<0.01),心肌CK、LDH含量明显升高(P<0.01);糖心乐大剂量组和格列齐特对照组大鼠FBS和心肌CK、LDH比较差异无统计学意义(P>0.05)。结论糖心乐对糖尿病心肌病大鼠有一定的降血糖作用,可减少糖尿病心肌病大鼠心肌酶漏出,纠正心肌能量代谢障碍。
Objective To investigate effects of Tangxinle(TXL) on blood sugar and myocardial enzyme in diabetic cardiomyopathy rats.Methods The model of diabetes mellitus was established with STZ by intraperitoneal injection in Sprague-Dawley rats,then diabetes mellitus rats were randomly divided into model control group,gliclazide control group,TXL high and low dose group,12 rats in each group.After eight weeks,the rats in gliclazide control group,TXL high and low dose group were separately perfused stomach with gliclazide(20 mg·kg-1),2.2 kg·L-1 TXL and 1.1 kg·L-1 TXL,the model control group was treated with normal saline,once a day.Ten normal rats were selected as normal control group.All treatments lasted four weeks,then fasting blood sugar(FBS) and activity of myocardial enzyme were detected.Results Four weeks after treatment,compared with the normal control group,the FBS increased,and the levels of CK,LDH in myocardial tissue were obviously decreased in model control group,gliclazide control group,TXL high and low dose group(P0.05).Conclusion TXL can reduce the level of plasma glucose,decrease leakage of myocardial enzyme from myocardial cell,improve the metabolic disorder of energy in diabetic cardiomyopathy rats.
