摘要 目的:探索白血病NB4细胞经全反式维甲酸(ATRA)诱导分化成熟后的中性粒细胞是否与正常中性粒细胞(PMN)之间存在差异,以了解急性早幼粒细胞白血病(APL)细胞的生物学特性。方法:ATRA诱导NB4细胞24、48、72、96 h后,瑞氏染色观察细胞分化情况、原子力显微镜(AFM)观察细胞表面地貌形态变化;NBT还原实验、吞噬杀菌功能实验和趋化功能实验研究细胞功能。结果:NB4细胞经ATRA诱导72 h后,细胞基本分化成熟,细胞核出现分叶,核/质比下降,与正常PMN相比,细胞直径相对较大,NBT还原率下降,趋化功能下降,但吞噬杀菌功能正常。AFM探测细胞机械性能发现,随着ATRA作用时间的延长,NB4细胞发生不同程度的形变,细胞体积增大,高度降低,均方根粗糙度(Rq)和平均粗糙度(Ra)降低。结论:ATRA诱导NB4分化成熟后的细胞和正常PMN,在形态和功能上存在一定差异。 Aim:To explore whether the ATRA induced NB4 is different from normal PMN and much better understand the biological characteristics of cells from APL patients, also to provide experimental ba- ses to understanding the rules of cell differentiation. Methods: Wright stain and AFM were used to ob- serve the morphology of NB4 cells which were induced by ATRA after 24 h, 48 h, 72 h, 96 h, NBT re- duction, bacteria killing ability and chemokinesis were also tested. Results: After 72 h, the induced NB4 cells were almost mature, the cells had folded, cleaved, or bilobed nuclei, and nucleocytoplasmic ra- tio decreased. Compared with PMN, NB4' s diameter was larger. There were also a tiny decreasing in NBT reduction and a slight defect in chemokinesis. Bacteria killing ability was normal. AFM result showed that the shape of NB4 cell was changing with the increasing of inducing time, such as increasing volume, reducing height, dropping mean roughness (Ra) and mean-square-root roughness (Rq). Conclu- sion:ATRA can induce NB4 into a mature one, there are some differences in the morphography and func- tion between normal PMN and induced NB4.
