离子对萃取光度法测定雪上一枝蒿总生物碱含量

Testing Total Content of Alkaloids from Radix Aconiti brachypodi by lon-pair Extration Spectrophotometry

目的 :解决雪上一枝蒿总生物碱含量测定。方法 :以雪上一枝蒿甲素为对照品 ,溴麝香草酚蓝为反离子 ,氯仿为萃取溶剂 ,建立了雪上一枝蒿药材、针剂中雪上一枝蒿总生物碱含量测定法。结果 :测定波长415± 0 5nm ,线性范围 2～ 10 μg·ml-1。方法平均回收率 ,纯品法 10 0 35± 1 12 (% ) ,RSD =1 12 % (n =9) ;加样法 98 5 5± 1 34 (% ) ,RSD =1 36 % (n =6 )。结论 :针剂中氯化钠不干扰测定。本法具有简便、迅速、准确之特点。

Objective: Test total content of alkaloids from Radix Aconiti brachypodi. Method: Used bullatin A (the formula C 22 H 33 O 2N,m343, mp 250～252℃, which has been proved to be identical with the known compound denudatine) as the contrasting standard, the extraction spectrophotometry with bromothymol blue as counterion had been established for the measurement of total content of alkaloids from Radix Aconiti Brachypodi or its injection. Result: The absorbance at 415±0 5nm showed the linear relationship in range of 2～10μg/ml with mean recovery of 100.35±1.12%, RSD=1.12%(n=9), (method Ⅰ-the contrasting standard), with that of 98.55±1.34%, RSD=1.36%(n=6), (method Ⅱ-put the liquid of control in the sample). Conclusion: The sodium chioride in injection showed no interference, the method is simple, rapid and accurate.
