摘要
以北方寒冷地区紫花苜蓿鲜草为原料,用纤维素酶破壁和机械破壁相结合提取叶蛋白。研究酶解反应温度,pH和加酶量三因素对苜蓿叶蛋白提取率的影响。在单因素试验基础上,通过响应面分析法确定纤维素酶法提取苜蓿叶蛋白的最佳工艺条件为:酶解反应温度为55.65℃、pH为4.86、加酶量为7.22%,在此工艺条件下,粗蛋白提取率为41.59%。通过氨基酸分析结果表明,苜蓿叶蛋白氨基酸种类齐全、含量高、氨基酸比例协调,因此苜蓿叶蛋白是优质蛋白质。
Alfalfa in northern cold areas as experiment material, leaf protein is extracted by combining cellulase with machine destroying cell wall. The effects of enzymolysis temperature, pH value and the quantity of enzyme on the extraction ratio of alfalfa leaf protein are investigated. On the basis of single-factor tests, the optimal conditions for cellulase extraction are achieved by response surface methodlogy, namely enzymolysis temperature 55.65 ℃, pH value 4.86 and the quantity of enzyme 7.22%. Under these technical conditions, extracting rate of protein is 41.59%. Amino acids analysis is employed, the result shows that the type of amino acids in leaf protein from alfalfa are complete, high content, and the proportion coordination of amino acids. Therefore, leaf protein from alfalfa is a kind of high quality protein.
