核仁蛋白PES1在乳腺癌细胞中的表达及siRNA干扰核仁蛋白PES1对乳腺癌细胞周期的影响

THE EXPRESSION OF NUCLEOLAR PROTEIN PES1 IN BCC AND THE EFFECT OF siRNA INTERFERED PES1 ON BCC CYCLE

目的探讨特异性siRNA干扰核仁蛋白PES1对乳腺癌细胞周期的影响。方法以Western Blot和免疫细胞化学法对PES1的表达和定位进行检测,用流式细胞仪检测细胞周期的变化。结果 PES1在细胞核内高表达,经siRNA干扰后,乳腺癌细胞G2期发生阻滞现象。结论核仁蛋白PES1在调控乳腺癌细胞周期中起着重要作用;siRNA干扰核仁蛋白PES1对乳腺癌细胞周期存在影响。

Objective To investigate the effect of nucleolar protein pescadillo-1 interfered with specific siRNA on the cell cycle of breast cancer.Methods The expression of PES1 was assessed by Western Blot and Immunohistochemical assay in breast cancer cell（BCC） lines,and Flow Cytometry was performed to detect the cell cycle.Results The results of the Western Blot and Immunohistochemical assay indicated that PES1 shown significant higher expression in breast cancer cell nuclear.Moreover,the cell BICR were arrested in G2 phase.Conclusion The nucleolar protein PES1 may play an important role in regulating the cell cycle of breast cancer,and have siRNA influence on it.
