摘要
目的探讨唐山地区IL-10基因启动子区-1082G/A多态性与汉族妇女HPV感染宫颈癌发生的相关性。方法应用PCR、ARMS-PCR技术对103例宫颈癌病例组和115例年龄匹配的正常对照组宫颈组织HR-HPV16、18DNA感染和IL-10基因启动子区域-1082多态性检测分析。结果 -1082G/A多态性GG、GA、AA基因型频率在HPV16阳性和HPV16阴性组中频率分别为:0、9.1%、90.9%和3.9%、19.2%、76.9%,差异无统计学意义(P>0.05);等位基因G和A在HPV16阳性和HPV16阴性组的频率分别为:4.5%、95.5%和13.5%、86.5%,其差异有统计学意义(P=0.049);IL-10基因启动子区-1082G/A多态性GG、GA、AA基因型频率在HPV18阳性和HPV18阴性组中分布分别为:0、21.1%、78.9%和1.2%、9.5%、89.3%,差异无统计学意义(P>0.05);等位基因G和A在HPV18阳性和HPV18阴性组的频率分别为:10.5%、89.5%和6.0%、94%,差异亦无统计学意义(P>0.05)。结论唐山地区汉族妇女IL-10基因启动子区-1082G/A多态性可能与高危型HPV16感染导致的宫颈癌有关。
Objective To investigate the association between single nucleotide polymorphism(SNP)-1082G/A sites within promoter region of IL-10 gene and HPV-associated cervical carcinoma in Han women in Tangshan city,Hebei province.Methods The distribution of HPV16/18 and the IL-10-1082G/A polymorphism of 101cases of cervical carcinoma and 100 cases of normal control were detected by using PCR,PCR-RFLP.Results-1082G/A polymorphism of GG,GA,AA genotype frequency in HPV16-positive and HPV16-negative group,frequencies were: 0,9.1%,90.9% and 3.9%,19.2%,76.9%,the difference was not obvious(P0.05);allele G and A in the HPV16 and HPV16 positive frequency of negative group were: 4.5%,95.5% and 13.5%,86.5%,a slight statistically significant the difference(P=0.049);IL-10 gene promoter-1082G/A polymorphism of GG,GA,AA genotypes HPV18 in HPV18-positive and-negative groups were distributed as follows: 0,21.1%,78.9% and 1.2%,9.5%,89.3%,the difference was not significant(P〉0.05);allele G and A in the HPV18 and HPV18 positive frequency of negative group were: 10.5%,89.5% and 6.0%,94%,differences were not significant(P〉0.05).Conclusion IL-10-1082G/A polymorphism may be associated with HPV16 infection in Han women in Tangshan area.
