摘要
利用RNAi技术,构建了AtPLC4基因的RNAi双元载体,并进行了拟南芥的遗传转化,通过抗性筛选及PCR鉴定,获得20株遗传转化株系.进一步通过RT-PCR检测,得到3株AtPLC4基因表达降低的转基因植株,为运用反向遗传学的方法深入研究AtPLC基因家族的生物学功能提供基础.
The RNAi vector for AtPLC4 gene was constructed and transformed into Arabidopsis. Twenty transgenic plants were identified by Hyg resistance screening and PCR. Three lines that lower expression of AtPLC4 gene were identified by RT-PCR. Those transgenic plants will be used to study the functions of the AtPLC gene family by the reverse genetics method.
