摘要
目的:研究生长激素释放激素受体剪接变体1(SV1),一种生长激素释放激素(GHRH)受体的变体在子宫内膜异位症的表达模式。方法:在位和异位子宫内膜组织、腹膜B细胞采自有或没有子宫内膜异位症的妇女,正常卵巢组织采自没有子宫内膜异位症的妇女。分离异位子宫内膜基质干细胞(ESC)并加入GHRH培养,GHRH和SV1在样品组织的基因表达由RT-PCR检测,BrdU的摄入由特定的分析系统检测。结果:54份子宫内膜异位组织中有34份(63.0%)表达SV1 mRNA,显著高于在位内膜组织(4.3%)和正常卵巢组织(0.0%)。GHRH mRNA在异位子宫内膜组织的表达为22.2%(12/54),在位内膜组织中为25.9%,而在正常卵巢组织中为100.0%(14/14)和腹膜B细胞为81.3%(13/16)。GHRH能刺激表达SV1的ESC增加BrdU摄入。结论:GHRH和SV1在异位内膜表达谱不同于在位内膜,GHRH激活SV1并刺激细胞的增殖。
Objective: To study the expression mode of growth hormone -releasing hormone (GHRH) receptor splice variant 1 ( SV1 ) , a kind of variant of GHRH, in endometriosis. Methods: The ectopic endometrial tissue samples and eutopie endometrial tissue sampies, peritoneal B cells were obtained from the women with and without endometriosis, and the normal ovarian tissue samples were obtained from the women without endometriosis. Ectopic endometrial stromal cells (ESC) were isolated, then they were cultured with GHRH. The gene expressions of GHRH and SV1 in the samples were detected by RT- PCR, the intake of 5 -bromo -2^- deoxyuridine (BrdU) was detected by special analytic system. Results. Among 54 samples of endometrial tissue, 34 samples (63.0%) expressed SV1 mRNA, which was statistically significantly higher than those in eutopic endometrium (4. 3% ) and normal ovarian tissue (0.0%) . The expression rates of GHRH mRNA in ectopic endometrium, eutopic endometrium, normal ovarian tissue, and peritoneal B cells were 22. 2% (12/54) , 25.9% , 100. 0% ( 14/14), and 81.3% ( 13/16), respectively. GHRH stimulated the intake of BrdU in ESC with expression of SV1. Conclusion: The expression patterns of GHRH and SV1 in ectopic endometrium are different from those in eutopie endometrium, GHRH can activate SV1 and stimulate cell proliferation.
