注射用海洋真菌多糖YCP血管刺激性、过敏性研究

Vascular irritation and anaphylaxis studies on preparaion of marine polysaccharide YCP

目的探索注射用海洋真菌多糖YCP在静脉给药后产生的血管刺激性以及豚鼠的全身主动过敏反应,为临床用药安全提供依据。方法血管刺激性试验:实验兔耳缘静脉给予18 mg/10(mL.kg)海洋真菌多糖YCP,每日1次,连续3 d,末次给药后48 h取兔耳进行病理组织学观察。全身主动过敏试验:豚鼠分别腹腔注射给予高、低剂量海洋真菌多糖YCP,隔日1次,共3次,末次致敏14 d后进行抗原激发,并观察豚鼠的全身过敏反应。结果血管刺激性试验期间各组兔耳注射部位未见异常改变;病理组织学检查未见异常。全身主动过敏试验致敏期间和激发后给药组和阴性组豚鼠无异常症状,阳性组豚鼠出现明显过敏症状。结论注射用海洋真菌多糖YCP无血管刺激性,全身主动过敏试验结果为阴性。

Objective To evaluate vascular irritation and anaphylaxis of marine polysaccharide YCP. Methods The vascular irritation of marine polysaccharide YCP for injection was injected to rabbit at the dose of 18 mg/kg, q.d. for 3 d. Pathology histology examine of injection place after stoping remedy 48 hours was made. Guinea-pigs were respectively sensitized 10 mg/kg and 5 mg/kg of marine polysaccharide YCP, 0.9% NaCl and ovalbumin by intraperitoneal injection, once every other day for 3 times. The guinea-pigs were injeceted antigens on the 14th day after the last sensitizing injection to evoke systemic anaphylaxis, signs were observed immediately after injection. Results No obvious irritation on rabbites＇ vascular and injection sites were noted. There was no abnormal symptoms of Guinea-pigs during sensitization period and evoking in administration groups and negative control group. Conclusion The vascular irritation and anaphylaxis are negative of marine polysaccharide YCP.
