西洛他唑对氧化低密度脂蛋白诱导的人单核细胞源树突状细胞成熟的影响

摘要 目的:研究西洛他唑对氧化低密度脂蛋白诱导的人单核细胞源树突状细胞成熟的影响。方法:分离正常人外周血单核细胞,rhGM-CSF和rhIL-4体外诱导分化为未成熟树突状细胞。随机分为对照组、氧化低密度脂蛋白(50μg/mL)组(刺激组)、西洛他唑(20μmol/L)预干预加氧化低密度脂蛋白(50μg/mL)组(干预组)。流式细胞仪检测细胞表型和内吞功能变化,酶联免疫检测细胞培养上清细胞因子水平。结果:与刺激组比较,干预组树突状细胞表面分子CD40和CD86的表达降低,内吞功能提高,细胞培养上清TNF-α和IL-6水平下降。结论:西洛他唑可抑制氧化低密度脂蛋白诱导的树突状细胞成熟,这可能参与了其抗动脉粥样硬化的机制。
