摘要
目的:探讨Neuropilin-1(NRP-1)mRNA在新生小鼠及促性腺激素预处理不同时间点小鼠卵巢中的表达。方法:选用C57BL/6j小鼠新生及促性腺激素预处理不同时间点卵巢,通过实时定量聚合酶链反应检测NRP-1mRNA表达。结果:NRP-1mRNA在新生第3天小鼠卵巢中的表达是第5天的1.7倍(P<0.05),是第14天的1.5倍(P<0.05);相对于对照生理盐水注射组(对照组),在孕马血清处理后4、8、12、24h,NRP-1mRNA表达显著降低(P<0.05),至24h最低,而48h显著增高为对照组的1.5倍(P<0.05);给予人绒毛膜促性腺激素后与0h相比NRP-1mRNA表达于第4小时下降至最低(P<0.05),后于第12小时升至最高(P<0.05)。结论:NRP-1在始基卵泡募集和卵泡发育、成熟及排卵过程中发挥重要作用。
Objective To explore the expression of neuropilin-1 (NRP-1 ) mRNA in the ovaries of newborn and gonadotropin-primed immature mice, Method Real-time polymerase chain reaction was used to detect the expression of NRP-1 mRNA in ovaries of newborn and gonadotropin-primed inmature C57BL/6j mice. Results The expression of NRP-1 mRNA in the ovaries of newborn C57BL/6j mice was significantly decreased in the ovaries of the 5 day group and the 14 day group compared to the 3 day group which was 1.7 times to the 5 day group and 1.5 times to the 14 day group(P 〈 0.05). The expression of NRP -1 mRNA diminished apparently as the PMSG treated till the 24 h to the lowest, and peaked at the 48h which was 1.5 times compared to the control group (P 〈 0.05 ). Then, as the addition of HCG to the treatment after 48 hours of PMSG, the expression of NRP-1 mRNA diminished to the lowest at the 4 hours and increased to the highest at the 12 hours (P 〈 0.05). Conclusion The NRP-1 may play an important role in the initial recruitment, follicle development, maturation and ovulation.
