摘要
目的探查甘丙肽能否促进大鼠脂肪细胞GluT4膜位移。方法用3 H葡萄糖测3T3L1脂肪细胞葡萄糖(2DG)摄取率;大鼠分4组:安静和运动对照组注射生理盐水,安静和运动用药组注射甘丙肽拮抗剂M35。两运动组大鼠游泳20d,60min/d。结果甘丙肽增加葡萄糖摄取;两用药组大鼠正糖钳的葡萄糖输注速率及脂肪细胞GluT4mRNA水平均下降,细胞外膜和总膜GluT4浓度降低,且外膜与总膜GluT4比值低于对照组。结论甘丙肽可促进GluT4膜转位而增强胰岛素敏感性。
Objective To explore the effect of galanin on translocation of glucose transporter 4(GluT4) in adipocytes of rats.Methods The glucose uptake rate was measured in 3T3 L1 adipocytes with 2-deoxy——D-glucose.Rats were randomized into 4 groups: the sedentary and exercise control groups were injected with saline,and the sedentary and exercise plus treatment groups were injected with M35,a galanin antagonist.Rats in both exercise groups swam for 20 d,60 min/d.Results Galanin treatment increased the glucose uptake rate,while M35 administration reduced glucose infusinon velocity in euglycemic-hyperinsulinemic clamp test,and decreased GluT4 mRNA expression and GluT4 concentration in plasma membranes and total cell membranes The ratio of GluT4 contents in plasma membranes to total cell membranes was lower in the treatment groups than in the control groups.Conclusion Galanin may promote GluT4 translocation to plasma membranes to enhance insulin sensitivity in adipocytes.
