摘要
用对酵母菌(YE4)有促进作用的乳酸菌(LC5)为研究对象,以OD值和活菌数为测定指标筛选合适的培养基。通过对5种乳酸菌常用培养基的筛选,确定M17培养基中LC5生长良好。以M17培养基作为基础培养基,进一步利用单因素试验和正交试验的设计方法对其进行优化,得出改良M17培养基的最佳成分为:大豆蛋白胨为0.6%,胰蛋白胨为0.6%,牛肉膏为0.4%,酵母浸提粉为0.5%,K2HPO4为0.7%,盐液A为0.4%,甘油为0.8%,乳糖为0.6%。优化后的培养基乳酸菌LC5的活菌数可达到1.89×109cfu/mL,是基础培养基的5.9倍,OD值是M17基础培养基的1.22倍。
We took lactic acid bacteria(LC5) as research object, which can promote the role of yeast(YE4), and took OD600 and viable count as determination indicators to filter suitable medium of LC5. Selected from 5 mediums which the LC5 usually used, result showed that the LC5 grows very well in M17 medium. So selected M17 as the base medium, by using single factor experiment and orthogonal test design method to optimize it, and the best ingredients of the modified M17 are that soy peptone 0.6%, tryptone 0.6%, beef powder 0.4%, yeast extract powder 0.5%, K2HPO4 0.7%, salt solution A 0.4%, glycerin 0.8%, and lactose 0.6%. The viable count of LC5 with modified M17 could reach 1.89x109 cfu/mL which are 5.9 times than basis medium with M17, also, the OD value is nearly 1.22 times than the basis M17 medium.
