摘要
以牛鼻软骨为原料酶法制备软骨粉,采用稀碱液浸提和乙醇沉淀的方法提取硫酸软骨素(Chondroitin Sulfate,ChS),通过单因素和正交试验对碱解工艺进行优化,确定的最佳条件是:料水比1:6,NaOH浓度5%,温度40℃,浸提时间6h,在此条件下,提取率达到12.58%,葡萄糖醛酸含量为32.44%,硫酸软骨素纯度达82%。紫外光谱分析:样品在196nm处有多糖的吸收峰,未见蛋白质(280nm)与核酸(260nm)的特征吸收峰;红外光谱分析发现为典型的硫酸软骨素A吸收峰。
The material powder was prepared from bovine nose cartilage by enzymatic hydrolysis method. ChS was extracted with alkaline solution and sedimentated with ethanol. The optimal alkaline hydrolysis conditions of cartilage powder were explored by single factor and orthogonal experiments to be the ratio of solid to liquid of 1:6, alkali solution of 5%, temperature of alkali extraction of 40 ~C and the duration of alkali extraction of 6 h. Under the optimal conditions, the yield was 12.58%, the content of Glucuronic acid was 32.44% and the purity quotient of Chondroitin Sulfate was 82%. UV analysis revealed that the absorption peak of polysaccharide appeared at 196 nm, while characteristic absorption peaks of protein (280 nm) and nucleic acid (260 nm) were not found. Typical absorption peaks of Chondroitin Sulfate A could be found by IR.
