多年生黑麦草MYB转录因子基因的克隆及序列分析 被引量:2

摘要 为进一步研究MYB类基因在多年生黑麦草抗逆和发育中的功能,以多年生黑麦草为材料,采用同源克隆的方法对其抗逆和发育相关MYB转录因子的保守区进行PCR扩增和克隆,并进行了序列分析和功能预测。测序结果表明,获得了3个不同的MYB转录因子基因片段,分别命名为LpMYB1、LpMYB2和LpMYB3。LpMYB1和LpMYB2长度均为221bp,内含子为103bp,编码39个氨基酸;LpMYB3长度为202bp,内含子为84bp,编码39个氨基酸;同源比对结果表明,所得3个MYB基因可能参与多年生黑麦草对逆境的应答和植株的形态建成;进化树分析表明,3个MYB基因的系统发育符合单子叶植物的进化模式。由此可见,3个MYB基因参与了多年生黑麦草抗逆性和发育的表达,为进一步研究多年生黑麦草MYB转录因子基因的功能研究奠定基础。 To further study the function of the MYB in stress resistance and development in Loli- urn perenne L. ,the conserved region of MYB transcription factor which is related with resilience and development was amplified by PCR using homologous method and cloned from leaves of the Lolium perenne L. Sequence analysis and function prediction were performed as well. Sequencing results showed that three different gene fragments of MYB transcription factor were cloned and named separately as LpMYB1 ,LpMYB2 and LpMYB3. The DNA lengths of LpMYB1 ,LpMYB2 and LpMYB3 were 221 bp,221 bp and 202 bp,which included 103 bp,103 bp and 84 bp introns, respectively. All of them encoded 39 amino acids. Sequence homologous comparison showed that these three genes might be involved in adversity response and morphogenesis of Loliurn perenne L. Phylogenetic analysis indicated that phylogenetic development of the three genes was consistent with the evolutionary model of monocotyledons. The results showed that the three MYB genes were involved in the expression of resistance and development of Lolium perenne L. , which provides a foundation for study of function of MYB transcription factor gene in Lolium perenne L.
