大鼠肠道缺血再灌注后肠粘膜损伤与防御素-5mRNA变化

摘要 目的探讨大鼠肠道缺血再灌注后肠粘膜损伤与大鼠防御素5(rat defensin-5,RD-5)mRNA变化。方法采用夹闭肠系膜上动脉(SMA)方法制造大鼠肠道缺血再灌注模型,大鼠分为假手术组、实验组,在缺血再灌注后1h、6h、12h及24h观测肠粘膜形态变化,检测肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)含量及大鼠防御素5(RD-5)mRNA的动态变化。结果形态学观察显示实验组缺血再灌注后肠粘膜呈不同程度的损害状态,损伤评分较假手术组高;实验组TNF-α、IL-6含量明显升高,12小时达高峰,均较假手术组高,并有统计学意义;大鼠防御素5(RD-5)mRNA在缺血再灌注后明显升高,再灌注后1小时到达高峰,随后逐渐下降,但均高于假手术组。结论肠道缺血再灌注对肠道粘膜造成损伤,肠道防御素5参与人肠粘膜的损伤与抗损伤过程。 Objective To investigate the intestinal mucosal injury and the change of rat defensin 5 mRNA after intestinal ischemia-reperfusion in rats. Method Clamping the superior mesenteric artery for 45 minutes and followed by reperiusion to manufacture rat intestinal ischemia-nperfusion model, the rats were divided randomly into sham operation 片roup and experimental group.After ischemia-reperfusion 1 h , 6 h, 12 h and 24 h, the blood samples and intestinal specimens were obtained to observe the intestinal morphological changes and detect the TNF-and alpha, IL-6 levels and the rats defension-5 (RD-5) mRNA. Results Morphological observations indicated that after ischemia-reperfusion the intestinal mucosa of the experimental group showed varying degrees oi 〈iarnage slate , and the damage scores were higher than lhe sham group. The serum levels of TNF-a and IL-6 oi the experimental group were statistically significantly increased when compared with the sham operation group and the 12 hours reached the peak. The rat defensin-5 (RD-5) mRNA was significantly inc^reased after ischemia and reperfusion,and reached the peak in the I hour group,then decreased gradually,Imt were still higher than the sham operation group. Conclusions The Intestinalischemia-reperfusion has damage to the intestinal mucosa, the intestinal defensin-5 may play an imporlanl role in nilestinal mucosal prolection and the repair process.
