摘要 利用DNA转化法将嗜胶质菌的功能基因转入嗜蜡菌中,成功构建一株嗜蜡、嗜胶质基因工程菌ZH20,分析此菌株的生物特性及高温下对原油的作用。结果表明:ZH20的生长特性与受体菌基本相同,最高耐受温度为75℃,对pH值的适应范围较广(4~10),对盐(NaCl)的最大耐受量为12.5%;基因工程菌ZH20不仅保留受体菌的嗜蜡性能,同时还获得供体菌的嗜胶质性能,能够有效降低原油凝固点和黏度。 A genetic engineering bacteria ZH2owas reconstructed by transforming genomic DNA from a colloid thirsty bacterium to a paraffin thirsty bacterium for enhancing biodegradation of paraffin and colloid. The biological characteristics of ZH20 and its effect on crude oil at high temperature were evaluated. The results show that the biological characteristics of ZH20 are analogous to the rec, ipient bacterium-the paraffin thirsty bacterium. ZH20 is stable at 75 ℃, acid or alkali stable at a pH range of 4-10, and NaCl stable at a concentration of 12.5%. The genetic engineering bacteria ZH20 inherits both paraffin thirsty and colloid thirsty characteristics, and it can simuhaneously and effectively reduce freezing point and viscosity of crude oil.
