扶正消癌方对Lewis肺癌模型小鼠的抑瘤作用 被引量:7

Tumor Inhibitory Effect of Fuzheng Xiaoai Formula on Lewis Lung Cancer M odel Mice

摘要 目的观察扶正消癌方对Lewis肺癌模型小鼠肿瘤生长的抑制作用及其机制。方法选择60只C57BL/6J小鼠右前肢腋下接种Lewis肺癌细胞建立Lewis肺癌小鼠模型,随机分为荷瘤模型组、环磷酰胺(CTX)组、中药预防组、综合组,每组15只。CTX组接种72h后一次性腹腔注射CTX溶液0.2ml(4mg/ml),中药预防组接种前5天开始灌胃扶正消癌方0.4ml(2g/ml),综合组在CTX组基础上灌胃扶正消癌方药0.4ml(2g/ml),荷瘤模型组给予0.4ml生理盐水灌胃;每天灌胃1次,至造模后12天。观察各组小鼠生存状态及移植瘤生长情况,称量瘤重、胸腺重,计算抑瘤率、胸腺指数。免疫组化法检测瘤组织细胞黏附因子CD44的表达。结果与荷瘤模型组比较,各组瘤重均显著降低(P<0.01),综合组瘤重显著低于CTX组(P<0.01),CTX组、中药预防组、综合组抑瘤率分别为83.97%、55.71%、87.23%。与荷瘤模型组比较,CTX组和综合组胸腺重均显著降低(P<0.05或P<0.01),CTX组胸腺指数显著降低(P<0.01);与CTX组比较,中药预防组胸腺重量显著增加(P<0.01),综合组胸腺指数显著增加(P<0.05)。综合组CD44表达程度最低,与CTX组比较差异有统计学意义(P<0.05)。结论扶正消癌方可以协同化疗药物,提高肿瘤细胞对免疫系统及抗癌药物的敏感性,下调或逆转Lewis肺癌模型小鼠黏附因子CD44表达,发挥抑瘤和抗转移作用。 Objective To observe the tumor inhibitory effect and its mechanism of Fuzheng Xiaoai Formula (formula for supporting the right-qi and resisting tumor) on Lewis lung cancer (LLC) mice. Methods Sixty C57BL/6J mice were used to establish LLC models and they were randomized into the cancer model group, cisplatin (CTX) group, Chinese medication group and combination group, with 15 in each. The CTX group was treated with intraperitoneal injection of 0.2ml (4mg/ml) CTX solution 72 hours after vaccination. The Chinese medication group was treated with intragastric administration of 0.4ml (2g/ml) Fuzheng Xiaoai Formula 5 days before modeling. The combination group was treated with both Fuzheng Xiaoai For- mula and CTX. The cancer model group was given 0.4ml of normal saline intragastrically. The intragastric administration re- peated once daily for 12 days. The survival status and the growth of transplanted tumors were observed. The weight of tumor and thymus was weighed. The tumor inhibitory rate and thymus index were calculated. The expression of CD44 was detected with immunohistochemical method. Results Comparing with the cancer model group, the tumor weight decreased significantly in other groups (P〈0.01). The tumor weight in the combination group was significantly lower than that in the CTX group (P d0.01). The tumor inhibitory rate of the CTX group, Chinese medication group and combination group was 83.97%, 55. 71% and 87.23 % respectively. Comparing with the cancer model group, the thymus weight in the CTX group and combination group decreased significantly (P〈0.05 or P〈0.01), and the thymus index decreased significantly in the CTX group (P〈 0.01). Comparing with the CTX group, the thymus weight in the Chinese medication group increased significantly (P〈0.01), and the thymus index in the combination group increased significantly (P〈0.05). The degree of CD44 expression in the combi- nation group was the lowest with statistical significance as compared with that in CTX group (P〈0.05). Conclusion Fuzheng Xiaoai Formula could increase the effectiveness of chemotherapy drugs, improve the sensitivity of tumor ceils to the immune system and anti-cancer drugs, down-regulating or reversing the expression of CD44 and play a role in tumor inhibitory and anti-metastasis.
