α-硫辛酸对Ⅱ型糖尿病大鼠心肌细胞凋亡的影响 被引量:2

摘要 目的探讨α-硫辛酸对Ⅱ型糖尿病心肌细胞凋亡的影响。方法 SD大鼠随机分为5组:正常对照组,糖尿病模型组,α-硫辛酸低,中,高量剂量组(n=8)。对照组大鼠给予普通饮食,实验组大鼠给予高脂高糖饮食1个月后腹腔注射小剂量的链尿佐菌素30 mg/kg,建立Ⅱ型糖尿病模型[1]。α-硫辛酸低,中,高量剂量组,并分别按15、30、60 mg/(kg.d)灌胃12周。检测血糖及血脂;测定超氧化物歧化酶(SOD)及谷光胱甘肽过氧化物酶(GSH-PX)的活性,原位末端转移酶标记技术(TdT-mediated dUTP nick end labeling,TUNEL)检测细胞凋亡。结果与对照组比较,糖尿病模型组大鼠血糖及甘油三脂(TG)、胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)明显增高(P<0.01),高密度脂蛋白胆固醇(HDL-C)明显降低(P<0.01),血清和心肌组织中的SOD,和GSH-PX的活性降低,心肌细胞凋亡率增加。与糖尿病模型组比较,α-硫辛酸干预组各指标得到改善。尤以中剂量组改善最明显,血糖及TG,TC,LDL-C明显降低(P<0.05),HDL-C显著提高(P<0.05);SOD和GSH-PX的活性有所改善,细胞凋亡率减少(P<0.05)。结论α-硫辛酸对Ⅱ型糖尿病大鼠心肌组织具有保护作用,其机制可能与其降低血糖,血脂水平,抑制氧化应激,进而降低细胞凋亡率有关。 Objective To investigate the effect of Alpha Lipoic Acid ( A - LA) on apoptosis of cardiomyocyte in II diabetic rat. Methods SD rats were randomly divided into five groups : control group, diabetic group, A - LA low, medi- um and high dose groups ( n = 8 ). Control group was given ordinary diet, and experimental group was given diet of high fat and sugar; after a month experimental group was injected small doses of streptozotoein to the intra -peritoneal of a urine at 30 mg/kg, thereby establishing type II diabetes model. A - LA low, medium and high dose groups were separately given 15, 30, 60 mg/ (kg . d) for 12 weeks. Then testing blood sugar and blood fat, the activities of SOD and GSH - PX were also measured with serum and heart tissue. Apoptosis was detected by TUNEL. Results Compared with control group, di- abetes model rats' blood sugar and TC, TC, and LDL - C increased significantly ( P 〈 0. 01 ) , while HDL - C reduced significantly ( P 〈 0. 01 ) , and the activities of SOD and GSH - PX were significantly reduced ( P 〈 0. 01 ) ; the rate of cell apoptosis was increased. Compared with the diabetic group, as for A - LA experimental group, all indexes above had been improved. The medium group's improvement is clear. Blood glucose and TG, TC, LDL - C reduced significantly ( P 〈 0. 05 ), HDL - C increased significantly ( P 〈 0. 05 ) , the activities of SOD and GSH - PX were improved ( P 〈 0.01 ) ; the rate of cell apoptosis was reduced. Conclusion Alpha Lipoie Acid has a protective effect on cardiac muscle in diabetic rats, the mechanism may be related to the inhibition of oxidative stress with low blood sugar and low blood fat level, and then the cell apoptosis rate is reduced .
