隐丹参酮诱导人肝癌HepG-2细胞凋亡作用研究 被引量:4

摘要 目的:探讨隐丹参酮诱导人肝癌细胞HepG-2细胞凋亡作用及机制。方法:采用MTT法测定隐丹参酮对人肝癌HepG-2细胞生长率的影响,流式细胞仪测定隐丹参酮诱导人肝癌细胞HepG-2细胞DNA的影响,蛋白印记测定隐丹参酮对人肝癌细胞HepG-2、caspase-3、PARP、bcl-2以及bax蛋白表达的影响。结果:隐丹参酮12.0μM-100μM对HepG-2细胞生长率的抑制与对照组比较具有显著性差异;隐丹参酮作用12h、24h和48h后,细胞凋亡率分别为1.95%、5.84%和9.05%;隐丹参酮(12μM)孵育24h和48h可显著促进caspse-3和PARP的裂解;隐丹参酮(12μM)孵育24h和48h可显著抑制bcl-2促进bax的蛋白表达。结论:隐丹参酮诱导人肝癌细胞HepG-2细胞凋亡,机制可能是通过抑制bcl-2,促进bax表达,通过调控bcl-2/bax比值,最终激活下游效应因子cas-pase-3,从而诱导细胞凋亡。
