玉米ZmAATP基因的克隆及遗传转化载体的构建 被引量:2

摘要 ATP-ADP转运蛋白(AATP)是质体内外能量供应和物质交换的重要载体,可以通过调节质体内外ATP浓度来影响质体内的合成代谢。本研究克隆了玉米ZmAATP基因,进化树分析显示ZmAATP蛋白与ATPase亲缘关系较近。进一步研发发现,授粉后随着玉米籽粒的发育和淀粉的积累,ZmAATP基因的表达量逐渐提高。另外,我们分别构建了由组成型启动子(Ubi)和胚乳特异型启动子(GluAI)驱动的ZmAATP基因的过量表达载体,为进一步研究该基因在玉米中的生物学功能奠定了基础。 ATP - ADP translocator protein (AATP) is an important carrier of energy supply and material exchange in plastids, which can regulate the ATP concentration both inside and outside of plastids to mediate constructive metabolism. In this study, we cloned a ZmAATP gene from maize. ZmAATP protein is phylogeneticly closer to ATPase. The expression levels of ZmAATP gene gradually increased in the maize grain after pollination. The expression cassettes of ZmAATP gene driven by the constitutive promoter (Ubi) or the endosperm - specific promoter (GluAI) were constructed, respectively.
