摘要 为优化齿瓣石斛无菌萌发条件,通过研究影响齿瓣石斛种子萌发的因子,探寻适合齿瓣石斛种子最优萌发的培养基及培养条件。结果表明:齿瓣石斛种子无菌培养的适宜培养基为MS培养基,蔗糖浓度为20g/L,浓度过低,萌发苗生长差,玻璃化严重;胚龄对齿瓣石斛的萌发率有重要影响,适宜的种子胚龄为10~11个月,胚龄时间短萌发率低;胚龄时间过长灭菌时蒴果易开裂,污染率大;添加0.3mg/L NAA,可缩短萌发时间,种子的萌发率、幼苗的成苗率和生长情况相比对照较好;不同培养条件萌发效果有明显的差异。适宜的萌发条件可促进齿瓣石斛萌发,提高萌发率,萌发后幼苗生长状况较好。 In order to optimize germination conditions for Dendrobiurn devonianum. Past,optimal media and culturing conditions for the seed germination were investigated by examining that factors that affect the seed germination. The results show that MS was the suitable sterile culture medium supplemented with 20 g/L sucrose. Lower sucrose concentration would result in poor germination seedling growth and serious vitrification. Embryo age exhibited significant impacts on the seed germination. Embryos with the age of 10 to 11 months were favorable for the seed germination. Lower germination rate appeared when the em- bryo age was short, while long embryo age would cause the crack of the capsule during sterilizing, and high pollution rate. Medium supplemented with 0.3 mg/L NAA, could shorten the germination time, and promoted the seed germination rate and seedling growth. Different culture conditions presented significant differences in seed germination, indicating the importance of selecting favorable germination conditions.
