高效液相色谱法测定忍冬藤中木犀草素的含量

Determination of Luteolin Content in CAULIS LONICERAE by HPLC

[目的]测定忍冬藤中木犀草素的含量。[方法]采用高效液相色谱法测定,色谱柱为Agilent ZORBAX SB-C18(4.6 mm×250 mm,5μm);流动相为无水甲醇-浓度0.3%磷酸水溶液(47∶53,V/V);流速为1 ml/min;柱温为30℃;检测波长为为350 nm。[结果]木犀草素的线性范围为0.5～2.5μg/ml,相关系数R=0.999 6,平均回收率为100.71%,RSD为1.9%;3批药材中木犀草素的平均含量为24.05μg/g。[结论]该方法简单、快速、可靠,适用于忍冬藤药材的质量控制。

[ Objective] To determine the luteolin content in CAULIS LONICERAE. [ Method] The HPLC method was performed on an Agilent ZORBAX SB- C18 （4.6 mm×250 mm,5μm）using an isocratic elutian with methanol-water containing 0.3% phosphoric acid （47： 53, WV） as the mobile phase at a flow rate of 1 ml/min ,the column temperature was 30 ℃ and the detection wavelength was set at 350 rim. [ Result] The linear rangs and regression equation of luteolin was 0.5 -2.5 μg/ml with R =0. 999 6. The average recovery was 100.71% ,RSD = 1.9%. The average luteolin content in the three batches of medicine was 24.05μg/g. [ Conclusion] This method is simple ,rapid and reliable for the quality control of CAULIS LONICERAE.
