摘要
目的观察眼针对急性脑缺血再灌注损伤模型大鼠脑组织细胞间黏附因子(ICAM-1)表达的影响。方法 64只SD大鼠随机分为4组:正常组(未处理)、假手术组(同模型组,仅鱼线插入深度1 cm)、模型组(应用线栓法复制脑缺血再灌注损伤大鼠模型,鱼线插入深度1.8~2 cm)和眼针组(模型成功后取肝区、上焦区、下焦区、肾区进行眼针治疗20 min,每日2次),每组16只。于再灌注后3 h进行神经功能缺损评分,采用免疫组化、Western印迹、实时荧光定量聚合酶链反应(RQ-PCR)方法测定脑组织ICAM-1蛋白及mRNA表达的变化。结果眼针治疗后可使大鼠神经功能缺损评分明显下降,并显著降低脑组织ICAM-1蛋白及mRNA表达(P<0.01)。结论眼针具有明显地改善大鼠脑缺血再灌注损伤的作用;其机制与下调脑组织ICAM-1表达有关。
Objective To investigate the effect of eye acupuncture therapy on hippocampus ICAM-1 expression in rats with acute cerebral ischemia-reperfusion(L/R) injury and the related mechanism, Methods The rat model of cerebral I/R was established by suture method. After reperfusion 3 h, the neurophysical behaviours were accessed by ZeaLonga neurophysical impairment marks. The methods of immunochemistry technique, Western-blot and RQ-PCR were taken to detect the alteration of the ICAM-1 protein and ICAM-1 mRNA expres- sions in rat hippocampus after the eye acupuncture therapy. Results Compared with that of model group, the neurologic impairment score of eye-acupuncture group was decreased obviously. The expressions of ICAM-1 protein and ICAM-1 mRNA in rat hippocampus after the eye ac- upuncture therapy were strongly decreased(P 〈 0. 05). Conclusions Eye acupuncture therapy can improve the cerebral I/R injury, evident- ly by decreasing the expression of the hippocampus ICAM-1 protein.
