摘要
目的观察宫颈鳞状细胞癌组织中Slug的表达情况,探讨其在宫颈鳞状细胞癌上皮-间充质转变(epithelial-mesenchymaltransition,EMT)中的作用机制。方法收集临床慢性宫颈炎标本25例、CIN 3级标本33例、宫颈鳞状细胞癌标本98例,分别用免疫组化、Western blot和实时荧光定量PCR检测Slug、E-cadherin、α-SMA蛋白及mRNA的表达情况。结果 Slug和α-SMA在宫颈鳞状细胞癌中高表达,并随恶性程度的升高而加强(P<0.01);E-cadherin在癌组织中的表达降低(P<0.01)。结论 Slug可能通过调控EMT从而在宫颈鳞状细胞癌的侵袭转移过程中发挥作用。
Purpose To investigate whether Slug regulated epithelial-mesenchymal transition of squamous cell carcinoma in cervix. Methods The protein and mRNA levels of Slug, E-cadherin and α-SMA were determined in 25 cases of chronic cervicitis 33 cases of cervical intraepithelial neoplasia and 98 cases of squamous cell carcinoma by immunohistochemistry, Western blot and real-time quanti- tative fluorescent PCR. Results That the protein and mRNA levels of Slug and c^-SMA in squamous cell carcinoma were higher than that of chronic cervicitis and cervical intraepithelial neoplasia tissues ( P 〈 0. 01 ). The expression of E-cadherin was decreased in squa- mous cell carcinoma (P 〈 0.01 ). Conclusions Slug plays a role in the process of invasion and metastasis by regulating epithelial- mesenchymal transition of squamous cell carcinoma in cervix.
