轴突导向因子EphA5沉默后抑制中脑神经上皮细胞投射

摘要 目的:探讨轴突导向因子EphA5在中脑神经上皮细胞向纹状体投射中的作用。方法:从孕11d大鼠胚胎取材并培养中脑神经上皮细胞;设计EphA5基因干扰序列,构建RNA干扰载体,采用电转染技术将EphA5干扰载体转入中脑神经上皮细胞,电转后将中脑神经上皮细胞与纹状体共培养,观察转染后中脑神经上皮细胞向纹状体组织的靶向生长情况。结果:中脑神经上皮细胞与纹状体组织块联合培养,神经突起明显朝向组织块生长;EphA5基因干扰载体转染中脑神经上皮细胞可降低EphA5蛋白表达,EphA5基因沉默后神经上皮细胞突起朝向纹状体生长趋势不明显。结论:EphA5基因沉默后中脑神经上皮细胞向纹状体投射减弱,EphA5基因可调控中脑神经上皮细胞向纹状体的靶向投射。 Objective: To explore the effects of EphA4 on the projection of mesocephalic neuroepithelial cells to the striatum. Methods: The mesocephalic neuroepitheliaI ceils from Wistar rat embryo at embryonic day 11 were cultured, and submitted to transfeet with recombinant plasmid containing RNA interference segments targeting EphA5. Then, the cells were co-cultured with the striatum, in order to observe the directional growth toward the striatum. Results: RNA interference caused knockdown of EphA5 expression. And, the transfected cells did not exhibit obvious directional growth toward the striatum. Conclusions: EphA5 may participate in regulation of projection from mesocephalie neuroepithelial cells to the striatum.
