摘要 目的:分离SCC-9细胞中CD44+的肿瘤细胞亚群,鉴定CD44+亚群细胞具有肿瘤干细胞的特性。方法:采用CD44免疫荧光抗体标记流式细胞术分选出CD44+的细胞亚群;检测各细胞亚群的细胞周期;检测CD133、Oct-4、BIM-1在CD44+和CD44-中的表达;检测各细胞亚群克隆形成率。结果:SCC-9细胞中CD44+的细胞亚群约占30%;CD44+的肿瘤细胞亚群70%处于G0/G1期;胚胎干细胞因子CD133、Oct-4、BIM-1在CD44+和CD44-的OSCCs中表达存在显著性差异(P<0.01);CD44+细胞亚群克隆形成率明显高于CD44-的细胞亚群。结论:在SCC-9舌鳞癌细胞系中CD44+细胞亚群具有肿瘤干细胞的特性。 Objective: To characterize the biological features of CD44+ cell populations in oral squamous cell carcinoma(OSCC), CD44+ and CD44 cell populations were sorted from SCC--9 line. Methods: To sort CD44+ subpopulation group of SCC--9 by flow cytometry. To detect the cell cycle of the CD44+ , CD44 cell populations. To detect the expression of CD133, Oct--4, BIM--1 gene and protein, compare gent and protein expression in the CD44+ cell population, CD44 cell population and ESCs. To detect cloning efficiency of the CD44+ cell population, CD44- cell population. Results: The results showed that a minority population of CD44+ cancer cells, which comprise about 30% of the cells in SCC--9. 70% CD44+cancer cells lied in G0/G1 period. CD44+ cells differentially expressed the CD133, Oct--4, BIM--1, ABCG2 genes, at both the RNA and protein levels, and the CD44+ cells in the tumor expressed high levels of these nuclear genes and protein(P〈0.01). Conclusion: CD44+ cells had the characteristics of cancer stem--like cells in lingua squamous cell carcinoma line SCC--9.
