摘要
目的:在体外观察温度升高对人牙周膜成纤维细胞活力的影响,探讨热牙胶根管充填的安全性。方法:将体外培养人牙周膜成纤维细胞悬液于不同温度及时间培养,用噻唑蓝(MTT)比色法、台酚蓝染色测定细胞的存活,流式细胞分析法测定人牙周膜成纤维细胞凋亡及死亡情况。结果:体外培养人牙周膜成纤维细胞的死亡率随着温度的上升逐渐增加。流式细胞分析显示:50℃组较对照组的坏死细胞比例明显上升,差异有统计学意义(P<0.01)。结论:温度升高能诱发人牙周膜成纤维细胞活力降低,死亡率、凋亡率上升。
Objective: To evaluate the effect of temperature rising on growth of human periodontal ligament fibroblasts. Methods: After cultured and passaged to fourth passage, the periodontal ligament cells were heated from 40℃to 50℃ and stayed 30 sec to 60 see at each time interval, respectively. Cells cultured at 37℃ 60 sec as control. The mortality rates of cells in different temperature groups were analysed by trypan--blue and the survival rates were evaluated by MTT (3 -- (4,5 -- dimethyhhiazol- 2 -- yl) -- 2,5 -- diphenyl tetrazolium bromide) eolorimetric method. The apoptosis rates were measured by flow cytometry. Results: The mortality rates were increased with the temperature rise. There were significant differences between 42℃ 30 sec and those higher groups with the con- trol (P^0.05). The survival rates were decreased with the temperature rise. The significant differences were found from 48℃ and 50℃ groups with the control (P〈0.01). And the rates of the cytoclasis cells were significantly in- creased when compared 50℃ 30 sec and 60 sec groups to 37℃ (P〈0.05) . Conclusion: Temperature rising could reduce survival and the metabolic capabilities of human periodontal ligament fibroblasts cells and might cause the cells cytoclasis.
