摘要
【目的】观察急性脑梗死(AcI)患者单个核细胞(MNCs)凋亡的变化,并检测周围血中细胞因子白细胞介素(IL)一1B和细胞间黏附分子-1(ICAM一1)水平的变化,以探讨细胞因子和黏附分子对MNCs凋亡的影响。【方法】采用流式细胞术检测58例AcI患者和30名健康对照者MNCs的凋亡情况,酶联免疫吸附法(ELISA)检测IL-I~和IcAM一1水平。【结果]ACI患者MNCs凋亡率明显高于健康对照组,随病情进展呈逐渐升高趋势;58例ACI患者在发病的72h内血清中IL一1B和IcAM一1显著高于健康对照组(P〈0.01),第1周达到最高值,至d。。呈逐渐下降趋势,且与MNCs凋亡率均呈正相关。【结论】ACI患者MNCs凋亡增加,可能与炎性细胞因子产生过多密切相关,通过适度调控细胞因子的分泌和MNCs凋亡有可能会改善ACI预后。
[Objective] To observe the change of mononuelear cell(MNCs) apoptosis in acute cerebral in- farction(ACI), and detect the levels of eytokines interleukin-ll3(IL-113) and intercellular adhesion molecule-1 (ICAM-1) in peripheral blood in order to explore the influence of eytokines and adhesion molecules in the ap- optosis of MNCs. [Methods] Tl^e apoptosis of MNCs in 58 ACI patients and 30 healthy controls were detected with flow cytometry. Enzyme-linked immunosorbent assay(ELISA) was used to examine the levels of IL-113 and ICAM-1. [Results] The apoptosis rate of MNCs in ACI patients with ACI was significantly higher than that in healthy controls, and had a gradually increasing trend. Serum levels of IL-1 i3 and ICAM-1 in 58 ACI patients within 72h of disease onset were significantly higher those in healthy controls( P d0. 01), and up to the peak value at 7 days, and then gradually declined at 14 days. The levels of IL-113 and ICAM-1 in ACI pa- tients were positively correlated to the apoptosis rate of MNCs. [Conclusion] The increasing of MNCs apopto- sis in ACI patients may be closely related to the over production of IL-113 and ICAM-1. The appropriate regula- tion of cytokine secretion and MNCs apoptosis may improve the prognosis of ACI.
