摘要
目的:对单壁碳纳米管(SWNTs)进行聚乙二醇(PEG)功能化修饰,考察修饰后产物的生物相容性。方法:对SWNTs进行共价及非共价PEG功能化修饰,利用红外光谱(IR)、和热失重分析(TG)进行表征。通过蛋白吸附试验和溶血试验评价碳纳米管无纺膜的生物相容性。结果:1-乙基-3-(3-二甲基氨丙基)-碳化二亚胺/N-羟基琥珀酰亚胺缩合法可以将PEG接枝到SWNTs上;利用吸附的性质可以将PEG非共价连接到SWNTs上。蛋白吸附试验结果表明,氧化单壁碳纳米管(o-SWNT)、PEG非共价修饰的SWNTs(n-PEG-SWNTs)和PEG共价修饰的SWNTs(c-PEG-SWNTs)的相平衡参数分别为49.7、4.0和3.1。溶血性试验结果表明,o-SWNTs、n-PEG-SWNTs和c-PEG-SWNTs的溶血率分别为(15.9±1.5)%、(1.6±0.6)%和(6.6±1.0)%。血小板黏附测试结果表明,PEG的引入明显抑制了血小板在材料表面的黏附。结论:PEG通过共价接枝或非共价吸附的方式连接到SWNTs上后可改善SWNTs的生物相容性。
OBJECTIVE Single-walled carbon nanotubes(SWNTs) was functionalized by polyethyleneglycol(PEG),the biocompatibility was investigated of after its functionalization.METHODS Infrared spectra(IR),thermogravimetric analysis(TG) were used to exosyndrome the SWNTs and functionalized SWNTs,protein adsorption test and hemolysis test were carried out to evaluate the biocompatibility of SWNTs nonwoven.RESULTS PEG would be covalently conjugated with SWNTs by EDC/NHS coupling reaction;PEG could also be noncovalently conjugated with SWNTs by its adsorption character.The equilibrium parameter of o-SWNTs,SWNTs adsorbed PEG noncovalently(n-PEG-SWNTs) and SWNTs grafted PEG covalently(c-PEG-SWNTs) in the protein adsorption test was 49.7,4.0 and 3.1 respectively.The hemolysis rate of o-SWNTs,n-PEG-SWNTs and c-PEG-SWNTs in hemolysis test was(15.9±1.5)%,(1.6±0.6)% and(6.6±1.0)% respectively.The platelet adhesion test experiments indicated that the introduction of PEG significantly inhibited the adhesion of platelets on the material surface.CONCLUSION Biocompatibility was improved after PEG covalently and noncovalently functionalization of single-walled carbon nanotubes.
