摘要
目的:研究青藤碱抑制P-糖蛋白(P-glycoprotein,P-gp)对小肠吸收更昔洛韦的影响。方法:采用小肠翻转试验和离体小肠吸收试验考察青藤碱促进更昔洛韦的吸收。结果:小肠翻转试验结果表明,经不同浓度的青藤碱溶液孵育后,更昔洛韦由小肠黏膜侧到浆膜侧的转运量随青藤碱浓度的增加而增加;离体小肠吸收试验结果表明,经不同浓度的青藤碱溶液孵育后,更昔洛韦由小肠浆膜侧到黏膜侧的转运量随青藤碱浓度的增加而减少。结论:青藤碱为P-gp抑制剂,有利于促进更昔洛韦的吸收。
OBJECTIVE To explore the enhancement of sinomenine on intestinal absorption of ganciclovir.METHODS The experiment of gut sac evertion and in vitro intestinal absorption tests were performed.ganciclovir was determined by HPLC.RESULTS The study of gut sac evertion showed that sinomenine significantly inhibited the function of intestinal P-glycoprotein(P-gp).The amount of ganciclovir transported from mucosa to serosa,after was increased if incubated with sinomenine solution or verapamil solution.The in vitro intestinal absorption test indicated that the transported ganciclovir from serosa to mucosa,incubated with sinomenine solution,was significantly decreased compared with control group.CONCLUSION Sinomenine can enhance ganciclovir intestinal absorption by inhibiting the function of P-gp.
